Brief description This proposal focuses upon understanding the structure and mechanism of perforin, a protein that performs a crucial role in human immunity. We will investigate how certain mutations in perforin cause deficiency and disease. Our work will also seek to understand how potential therapeutics act to modulate perforin function. Finally, through a combination of biochemical and structural studies we will elucidate the details of perforin mechanism.
