The abundance, biomass, and diversity of microzooplankton and their herbivorous impact on phytoplankton were examined on a cross shelf transect (see thumbnail), sampling five stations from February 2002 to December 2004, and in a pair of mesoscale eddies ~300km from the coast on the seaward side of the Leeuwin Current. Prior to this study, no research on microzooplankton from temperate waters off Western Australia had been undertaken.Lineage

Statement: Prior to each cruise, 100 mL of acid Lugol's fixture was added to 1 L sample bottles and each bottle was weighed; they were reweighed after the cruise to determine the amount of sea water collected.
Water samples were collected from 10 or 5 L Niskin bottles deployed on a rosette fitted with a CTD (Seabird Model SBE 19+). Salinity, temperature, chlorophyll a, and photosynthetically available radiation (PAR) were measured.
A silicon tube, fitted to the spigot, was used to drain water from the Niskin to the sample bottle. The sea water was added just below the level of the Lugol's to ensure that cells were fixed at the highest possible concentration of Lugol's. The samples were stored in a cool,dark environment and analysed within 2 months of collection.
The volume of samples were reduced by settling and siphoning. After reweighing, the samples were settled and decreased to 250 mL by siphoning off the supernatant through a 10 um mesh cover. The samples were resettled in a tall thin tube and again siphoned down to 50 mL by the same method as above. The sample was poured into an Utermohl tube and settle for at least 24 hours. Ten percent of the coverslip area was examined, and every heterotrophic/mixotrophic organism between 20 - 200 um and all protists >200 um were counted with a Leica (DM IRB) inverted microscope at X200 magnification using a calibrated optical micrometer.
