Cytiva Akta Pure 25 Liquid Chromatography with Fraction Collector.The Cytiva AKTA pure 25 Liquid Chromatography System with Fraction Collector is a modular, research-scale liquid chromatography platform used for the automated purification of proteins, peptides, and nucleic acids. The principle of chromatography is based on the differential interaction of sample components with a stationary phase packed in a chromatography column and a liquid mobile phase, enabling separation of biomolecules according to physicochemical properties such as size, charge, hydrophobicity, or specific binding affinity. The AKTA pure 25 system accommodates a broad range of Cytiva laboratory-scale chromatography columns and column formats, including HiTrap, HiScreen, HiPrep, HiLoad, RESOURCE, Tricorn, and Precision columns, provided that column operating limits are compatible with system flow and pressure conditions. Compatible chromatography applications include size exclusion chromatography (SEC), desalting/buffer exchange, ion exchange chromatography (IEX), affinity chromatography, hydrophobic interaction chromatography (HIC), multimodal chromatography (MMC), and reversed phase chromatography (RPC). Within the system, integrated pumps, mixers, valves, and monitors are used to control buffer delivery, sample loading, column equilibration, washing, gradient elution, and peak detection during purification runs. The AKTA pure 25 configuration supports flow rates of up to 25 mL/min and is intended for laboratory-scale purification workflows ranging from microgram quantities to tens of grams of target biomolecule. The system includes UV detection, conductivity monitoring, and pressure monitoring for real-time observation of chromatographic performance, while the associated fraction collector enables automated collection of eluted fractions according to the programmed method. The instrument is operated through UNICORN software, which is used for method creation, system control, run monitoring, and data management, allowing standardized and reproducible execution of chromatography purification procedures in research laboratory settings.
