Chapter 3 presents a manipulative field experiment at Herdsman Lake, an urban wetland in metropolitan Perth, examining the microbial communities that colonise plastic buried in sediment and whether they carry the genetic capacity to degrade it. Low-density polyethylene (LDPE) films were deployed as the test substrate alongside polytetrafluoroethylene (PTFE) as an inert negative control, buried for 90 days and retrieved across a time series together with paired bulk sediment samples. Genomic DNA was extracted from plastic-associated biofilms and sediment and sequenced by shotgun metagenomics.
